Golgi-Cox staining, also known as Golgi-Cox impregnation, is one of the most elegant and effective procedures for studying the morphology of neurons and glia. It remains a primary technique for visualizing dendritic branching patterns and dendritic spines, allowing isolation and visualization of dendritic arbours from a small random fraction of neurons in a given brain area. Golgi techniques are widely used not only for anatomical studies but also in research examining behavioral-morphological relationships. Applications include identifying autonomic innervations in the heart and peripheral tissues.
Traditional Golgi-Cox staining is time-consuming, requires a vibratome, yields relatively few stained cells, and often produces inconsistent results. The Hito Golgi-Cox OptimStain™ Kit, designed based on methods by Glaser and Van der Loos, provides significant improvements. Procedures are simplified, processing time is reduced, and staining quality is stable and reliable with minimal overstaining and artifacts.
Users can employ a cryostat set at –19ºC for sectioning, or alternatively prepare paraffin sections with a microtome. The Standard Kit supports up to 50 mouse brains.
