The Perl’s stain is the most widely used method for detecting iron in biopsy specimens. It relies on the Prussian blue reaction. Historically introduced in 1867 by German pathologist Max Perls (1843–1881), the original formula used separate solutions of potassium ferrocyanide and acid to stain tissue. In this process, tissue is first treated with dilute hydrochloric acid to release ferric ions from binding proteins. The freed ions then react with potassium ferrocyanide to form insoluble Prussian blue dye (hydrated ferric ferrocyanide) in situ.
