Von Kossa staining has been used since the 19th century to demonstrate calcium deposits in tissues. While not specific for calcium itself, the method stains calcium salts such as calcium phosphate and calcium carbonate. Silver ions in the staining solution bind with phosphate or carbonate, replacing calcium in a precipitation reaction. Under strong light illumination, the silver salts are reduced to black metallic silver, allowing indirect visualization of calcium deposits in tissue samples.
The Hito Von Kossa OptimStain™ Kit produces brown to black calcium salts against red cytoplasm and nuclei in just 90 minutes. The kit includes a developer solution, eliminating the need for exposure to strong light, sunlight, or ultraviolet light. This ensures stable and improved staining quality.
For verification of staining results, Calcium Positive Control Slides are included (16–18 day mouse embryo).
